Global warming leads to the melting of permafrost, affects changes in soil microbial community structures and related functions, and contributes to the soil carbon cycle in permafrost areas. Located at the southern edge of Eurasia's permafrost region, the Greater Khingan Mountains are very sensitive to climate change. Therefore, by analyzing the bacterial community structure, diversity characteristics, and driving factors of soil profiles (active surface layer, active deep layer, transition layer, and permafrost layer) in this discontinuous permafrost region, this research provides support for the study of the carbon cycling process in permafrost regions. The results show that the microbial diversity (Shannon index (4.81)) was the highest at 0-20 cm, and the Shannon index of the surface soil of the active layer was significantly higher than that of the other soil layers. Acidobacteria and Proteobacteria were the dominant bacteria in the active layer soil of the permafrost area, and Chloroflexi, Actinobacteria, and Firmicutes were the dominant bacteria in the permafrost layer. Chloroflexi made the greatest contribution to the bacterial community in the permafrost soil, and Bacteroidota made the greatest contribution to the bacterial community in the active surface soil. The structure, richness, and diversity of the soil bacterial community significantly differed between the active layer and the permafrost layer. The number of bacterial species was the highest in the active layer surface soil and the active layer bottom soil. The difference in the structure of the bacterial community in the permafrost soil was mainly caused by changes in electrical conductivity and soil-water content, while that in the active layer soil was mainly affected by pH and soil nutrient indices. Soil temperature, NO3--N, and pH had significant effects on the structure of the bacterial community. The active layer and permafrost soils were susceptible to environmental information processing and genetic information processing. Infectious disease: the number of bacterial species was significantly higher in the surface and permafrost layers than in the other layers of the soil. In conclusion, changes in the microbial community structure in soil profiles in discontinuous permafrost areas sensitive to climate change are the key to soil carbon cycle research.
2024-08-01 Web of ScienceUnder the background of climate change, freeze-thaw patterns tend to be turbulent: ecosystem function processes and their mutual feedback mechanisms with microorganisms in sensitive areas around the world are currently a hot topic of research. We studied changes of soil properties in alpine wetlands located in arid areas of Central Asia during the seasonal freeze-thaw period (which included an initial freezing period, a deep freezing period, and a thawing period), and analyzed changes in soil bacterial community diversity, structure, network in different stages with the help of high-throughput sequencing technology. The results showed that the alpha diversity of the soil bacterial community showed a continuous decreasing trend during the seasonal freeze-thaw period. The relative abundance of dominant bacterial groups (Proteobacteria (39.04%-41.28%) and Bacteroidota (14.61%-20.12%)) did not change significantly during the freeze-thaw period. At the genus level, different genera belonging to the same phylum dominated in different stages, or there were clusters of genera belonging to different phylum. For example, g_Ellin6067, g_unclassified_f_Geobacteraceae, g_unclassified_f_Gemmatimonadaceae coexisted in the same cluster, belonging to Proteobacteria, Desulfobacterota and Gemmatimonadota respectively, and their abundance increased significantly during the freezing period. This adaptive freeze-thaw phylogenetic model suggests a heterogeneous stress resistance of bacteria during the freeze-thaw period. In addition, network analysis showed that, although the bacterial network was affected to some extent by environmental changes during the initial freezing period and its recovery in the thawing period lagged behind, the network complexity and stability did not change much as a whole. Our results prove that soil bacterial communities in alpine wetlands are highly resistant and adaptive to seasonal freeze-thaw conditions. As far as we know, compared with short-term freeze-thaw cycles research, this is the first study examining the influence of seasonal freeze-thaw on soil bacterial communities in alpine wetlands. Overall, our findings provide a solid base for further investigations of biogeochemical cycle processes under future climate change.
2023-12-01 Web of ScienceHigh latitude regions are experiencing considerable winter climate change, and reduced snowpack will likely affect soil microbial communities and their function, ultimately altering microbial-mediated biogeochemical cycles. However, the current knowledge on the responses of soil microorganisms to snow cover changes in permafrost ecosystems remains limited. Here, we conducted a 2-year (six periods) snow manipulation experi-ment comprising ambient snow and snow removal treatments with three replications of each treatment to explore the immediate and legacy effects of snow removal on soil bacterial community and enzyme activity in secondary Betala platyphylla forests in the permafrost region of the Daxing'an Mountains. Generally, bacterial community diversity was not particularly sensitive to the snow removal. Seasonal fluctuations in the relative abundance of dominated bacterial taxa were observed, but snow removal merely exerted a significant impact on the bacterial community structure during the snow melting period and early vegetation growing season within two consecutive years, with a reduction in the relative abundance of Chloroflexi and an increase in the relative abundance of Actinobacteria, and no evidence of cross-season legacy effects was found. Moreover, snow removal significantly altered the soil enzyme activities in the snow stabilization period and snow melting period, with an increase in soil acid phosphatase (ACP) activity of snow melting period and a decrease in polyphenol oxidase (PPO) activity of snow stabilization period as well as beta-glucosidase (BG) activity of snow stabilization period and snow melting period, but this effect did not persist into the vegetation growing periods. The seasonal variations in bacterial community and enzyme activity were mostly driven by changes in soil nutrient availability. Overall, our results suggest that soil bacterial communities have rather high resilience and rapid adaptability to snow cover changes in the forest ecosystems in the cold region of the Daxing'an Mountains.
2023-11-01 Web of ScienceLimited knowledge about the variation patterns of bacterial community composition in the sand and vegetative ecosystems confines our understanding regarding the contribution of the sand dune to desert areas. In this study, 454 pyrosequencing platforms were adopted to determine the community structure of bacteria and diversity of sand dunes in northeastern Qinghai-Tibet Plateau, China: 50 cm deep, rhizosphere, physical crusts, and biological crusts representing sand and vegetative ecosystems, respectively. The findings revealed significant variation in bacterial diversities and the structure of communities in the sand and vegetative ecosystems. The dominant bacterial phyla of sand and vegetative ecosystems were Firmicutes (47%), Actinobacteria (21%), Proteobacteria (16%), and Bacteroidetes (13%), while Lactococcus (50%) was found to be the dominant genus. Furthermore, samples with high alpha-diversity indices (Chao 1 and Shannon) for the vegetative ecosystem have the lowest modularity index and the largest number of biomarkers, with some exceptions. Redundancy analysis exhibited that environmental factors could explain 72% (phyla) and 67% (genera) of the bacterial communities, with EC, TC, and TOC being the major driving factors. This study expands our understanding of bacterial community composition in the desert ecosystem. The findings suggest that variations in the sand and vegetative ecosystems, such as those predicted by environmental factors, may reduce the abundance and diversity of bacteria, a response that likely affects the provision of key ecosystem processes by desert regions.
2022-12Climate warming can affect freeze-thaw cycle (FTCs) patterns in northern high-latitude regions and may affect permafrost carbon emissions. The response of carbon release and microbial communities to FTCs has not been well characterized. Here, we conducted laboratory incubation experiments to investigate the relationships among carbon emissions, bacterial community, and soil variables in a permafrost-affected forest-wetland ecotone in Northeast China. The emission rates of CO2 and CH4 increased during the FTCs. FTC amplitude, FTC frequency, and patch type had significant effects on carbon emissions. FTCs increased the contents of soil DOC, NH4+-N, and NO3--N but reduced bacterial alpha diversity. CO2 emissions were mainly affected by bacterial alpha diversity and composition, and the inorganic nitrogen content was the important factor affecting CH4 emissions. Our findings indicated that FTCs could significantly regulate CO2 and CH4 emissions by reducing bacterial community diversity and increasing the concentration of available soil substrates. Our findings shed new light on the microorganism-substrate mechanisms regulating the response patterns of the soil carbon cycle to FTCs in permafrost regions.
2022-10-01 Web of ScienceIn Arctic soils, warming accelerates decomposition of organic matter and increases emission of greenhouse gases (GHGs), contributing to a positive feedback to climate change. Although microorganisms play a key role in the processes between decomposition of organic matter and GHGs emission, the effects of warming on temporal responses of microbial activity are still elusive. In this study, treatments of warming and precipitation were conducted from 2012 to 2018 in Cambridge Bay, Canada. Soils of organic and mineral layers were collected monthly from June to September in 2018 and analyzed for extracellular enzyme activities and bacterial community structures. The activity of hydrolases was the highest in June and decreased thereafter over summer in both organic and mineral layers. Bacterial community structures changed gradually over summer, and the responses were distinct depending on soil layers and environmental factors; water content and soil temperature affected the shift of bacterial community structures in both layers, whereas bacterial abundance, dissolved organic carbon, and inorganic nitrogen did so in the organic layer only. The activity of hydrolases and bacterial community structures did not differ significantly among treatments but among months. Our results demonstrate that temporal variations may control extracellular enzyme activities and microbial community structure rather than the small effect of warming over a long period in high Arctic soil. Although the effects of the treatments on microbial activity were minor, our study provides insight that microbial activity may increase due to an increase in carbon availability, if the growing season is prolonged in the Arctic.
2022-07-01 Web of ScienceBackground Soil microorganisms in the thawing permafrost play key roles in the maintenance of ecosystem function and regulation of biogeochemical cycles. However, our knowledge of patterns and drivers of permafrost microbial communities is limited in northeastern China. Therefore, we investigated the community structure of soil bacteria in the active, transition and permafrost layers based on 90 soil samples collected from 10 sites across the continuous permafrost region using high-throughput Illumina sequencing. Results Proteobacteria (31.59%), Acidobacteria (18.63%), Bacteroidetes (9.74%), Chloroflexi (7.01%) and Actinobacteria (6.92%) were the predominant phyla of the bacterial community in all soil layers; however, the relative abundances of the dominant bacterial taxa varied with soil depth. The bacterial community alpha-diversity based on the Shannon index and the phylogenetic diversity index both decreased significantly with depth across the transition from active layer to permafrost layer. Nonmetric multidimensional scaling analysis and permutation multivariate analysis of variance revealed that microbial community structures were significantly different among layers. Redundancy analysis and Spearman's correlation analysis showed that soil properties differed between layers such as soil nutrient content, temperature and moisture mainly drove the differentiation of bacterial communities. Conclusions Our results revealed significant differences in bacterial composition and diversity among soil layers. Our findings suggest that the heterogeneous environmental conditions between the three soil horizons had strong influences on microbial niche differentiation and further explained the variability of soil bacterial community structures. This effort to profile the vertical distribution of bacterial communities may enable better evaluations of changes in microbial dynamics in response to permafrost thaw, which would be beneficial to ecological conservation of permafrost ecosystems.
2022-01-11 Web of ScienceAs a buffer layer for the energy and water exchange between atmosphere and permafrost, the active layer is sensitive to climate warming. Changes in the thermal state in active layer can alter soil organic carbon (SOC) dynamics. It is critical to identify the response of soil microbial communities to warming to better predict the regional carbon cycle under the background of global warming. Here, the active layer soils collected from a wetland-forest ecotone in the continuous permafrost region of Northeastern China were incubated at 5 and 15 degrees C for 45 days. High-throughput sequencing of the 16S rRNA gene was used to examine the response of bacterial community structure to experimental warming. A total of 4148 OTUs were identified, which followed the order 15 degrees C > 5 degrees C > pre-incubated. Incubation temperature, soil layer and their interaction have significant effects on bacterial alpha diversity (Chao index). Bacterial communities under different temperature were clearly distinguished. Chloroflexi, Actinobacteria, Proteobacteria, and Acidobacteria accounted for more than 80% of the community abundance at the phylum level. Warming decreased the relative abundance of Chloroflexi and Acidobacteria, while Actinobacteria and Proteobacteria exhibited increasing trend. At family level, the abundance of norank_o__norank_c__AD3 and Ktedonobacteraceae decreased significantly with the increase of temperature, while Micrococcaccac increased. In addition, the amount of SOC mineralization were positively correlated with the relative abundances of most bacterial phyla and SOC content. SOC content was positively correlated with the relative abundance of most bacterial phyla. Results indicate that the SOC content was the primary explanatory variable and driver of microbial regulation for SOC mineralization. Our results provide a new perspective for understanding the microbial mechanisms that accelerates SOC decomposition under warming conditions in the forest-wetland ecotone of permafrost region.
2021-12-24 Web of ScienceDesert soil around the black stones is highly complex, which substantially affects the diversity and composition of inhabiting microbes. The existence of black stones in the southern part of the Black Gobi desert of China could provide microhabitats for diverse bacterial communities that remain unexplored. Hence, Illumina MiSeq sequencing was used to determine the differences in bacterial communities associated within microhabitats in three sites of the Black Gobi desert, China. Our results show that bacterial communities are significantly affected by each microhabitat. For instance, the a-diversity of bacterial communities indicated more remarkable diversity and richness in these microhabitats. Considering beta-diversity, variances were reported mainly in the Proteobacteria (30%), Actinobacteria (26%), Chloroflexi (19%), and Firmicutes (9%). Firmicutes were markedly enriched in the upper surface, especially in site 1. Compared to other microhabitats, the relative abundance of Proteobacteria was greater in the subsurface, and they were also more dominant in the other two sites. Network analysis of soil factors and bacterial genera showed that the most significant-occurrences were positively correlated, demonstrating potential synergistic interactions. Collective with the predicted function profiles and the redundancy analysis, these results indicated the highest variances in bacterial community structure and function in Black Gobi Desert ecosystems. These differences are likely closely related to the soil parameters, mainly water content, total carbon, and total nitrogen, and might be associated with black stones. This study concludes that microhabitats formed by black stones support highly diverse and biologically active bacterial communities. These microhabitats with extreme environmental conditions deliver new opportunities to explore soil bacterial communities at relevant spatial scales in the Black Gobi desert.
2021-12Boreal forests in permafrost zone store significant quantities of carbon that are readily threatened by increases in fire frequency and temperature due to climate change. Soil carbon is primarily released by microbial decomposition that is sensitive to environmental conditions. Under increasing disturbances of wildfire, there is a pressing need to understand interactions between wildfires and microbial communities, thereby to predict soil carbon dynamics. Using Illumina MiSeq sequencing of bacterial 16S rDNA and GeoChip 5.0K, we compared bacterial communities and their potential functions at surface and near-surface permafrost layers across a chronosequence ( > 100 years) of burned forests in a continuous permafrost zone. Postfire soils in the Yukon and the Northwest Territories, Canada, showed a marked increase in active layer thickness. Our results showed that soil bacterial community compositions and potential functions altered in 3-year postfire forest (Fire(3)) comparing to the unburned forests. The relative abundance of Ktedonobacteria (Chloroflexi) was higher in Fire(3) surface soils, while Alphaproteobacteria and Betaproteobacteria (Proteobacteria) were more abundant in unburned ones. Approximately 37% of the variation in community composition can be explained by abiotic variables, whereas only 2% by biotic variables. Potential functional genes, particularly for carbon degradation and anammox, appeared more frequent in Fire 3 than in unburned soils. Variations in functional gene pools were mainly driven by environmental factors (39%) and bacterial communities (20%; at phylum level). Unexpectedly, wildfire solely altered bacterial communities and their functional potentials of the surface layers, not the near-permafrost layers. Overall, the response of bacterial community compositions and functions to wildfire and the environment provides insights to re-evaluate the role of bacteria in decomposition.
